pH-Wert. 7,2 ± 0,2 HERSTELLUNG 31,6 g des Mediums werden in einem Liter destillierten Wasser suspendiert. Man mische gut, um eine einheitliche Suspension zu erhalten. Man erhitze unter häufigem Rühren/Schütteln und lasse eine Minute lang kochen. Man sterilisiere 15 Minuten lang im Autoklaven bei 118 -121 °C. EINSATZGEBIET Bengalrot-Agar (mit Chloramphenicol) ist ein Selektivmedium für Pilze und Hefen in Lebensmitteln. Der hohe Anteil Bengalrot in der Formulierung inhibiert übermäßiges Wachstum schnell wachsender Pilze, so dass langsamer wachsende Spezies nicht überwuchert werden. Hefen erscheinen durch das Bengalrot rosa gefärbt. Chloramphenicol hemmt bakterielles Wachstum. Die Platten können angeimpft werden, indem man 0,1 ml der verdünnten Probe auf der fertigen Platte ausstreicht. Alternativ hierzu kann auch angeimpft werden durch die „Gieß-Methode“, bei der man das autoklavierte Medium auf 45 °C abkü hlen lässt, dann 1 ml der verdünnten Probe in eine leere Petrischale gibt und sofort flüssiges Medium dazugießt. Die Inkubation erfolgt bei 22 °C für ca. 5 Tage. MIKROBIOLOGISCHE TESTS Die folgenden Ergebnisse wurden erzielt nach Inkubation von Referenzstämmen im angegebenen Medium / Agar bei einer Temperatur von 25 °C für 5 Tage.

Approximate formula in g/l: Peptone .5.0 Dextrose .10.0 Potassium phoshphate.1.0 Magensium sulfate .0.5 Chloroamphenicol .0.1 Rose Bengal .0.05 Agar .15.0
PREPARATION Suspend 31.6 g of the medium in one liter of distilled or deionized water. Mix well to obtain an even uniform suspension. Heat with frequent agitation and boil for one minute. Dispense and sterilize at 118-121 °C for 15 minutes. USES Rose Bengal-agar (with chloroamphenicol) is a selective media for fungi and yeast in food. The high amount of Rose Bengal in the formulation inhibits excessive growth of fast-growing fungi, so that slow-growing species are not overgrown. Yeast colonies may have a pink appearance caused by the Rose Bengal. Chloroamphenicol inhibits bacterial growth. The plates can be inocculated by plating 0.1 ml diluted sample onto the polymerized agar plate. Alternatively you can also inocculate by using the ‘casting method’, i.e. allow the autoclaved media to cool down to 45 °C, then put 1 ml diluted sample into an empty Petri dish and add liquid media immediately. Incubate for approx. 5 days at 22 °C.
MICROBIOLOGICAL TEST The following results were obtained in the performance of the given medium from type cultures after incubation at a temperature of 25 °C and observed a fter 5 days.
